数万年前，神魔大战，源生之神泰和利用世间至高至强的神器“神之左手”封印魔神，结果神器折毁坠落人界，源生之众神陷人长眠，自此三界再无神迹。战鬼族趁乱崛起，引发大战，神女无双受天界之托，化身人族少女进入神仆家族有狐一族的洞府，成为祭司源仲的贴身侍女，计划夺回他体内寄生已久的神之左手。但无双却看到本可用神之左手反抗的源仲被族人孤立、诋毁，她为之心疼为之可怜，决定保护源仲帮助他解开封印，与他一起开心的度过生命中所剩不多的时光。但死而复生的战鬼族陡然凯旋，在有狐族内部引起恐慌。早已被族人迫害濒临堕魔的源仲借机下山解开封印，但下山途中又被无双所治愈，两个人互诉心意情投意合。最终源仲救出被情敌迫害身陷禁锢的无双，两人联手合力打败了韩女，为三界众生换来了太平秩序。

本作品讲述了在警察“机动搜查队”，机动能力和运动神经首屈一指、但却没有刑事搜查经验也缺乏刑警常识的伊吹蓝（绫野刚饰），观察和社交能力突出,但却不信任自己和别人的理性的刑警、志摩一未（星野源饰），主角二人以24小时的时限内破案为目标的故事。标题中的“MIU”是MobileInvestigativeUnit（机动搜查队）的首字母，“404”是两人饰演机动搜查队员的呼叫信号。
